Strict Criteria Sperm Morphology for Infertility Diagnosis and Treatment, Semen
Use
This test is useful for diagnosing male infertility and selecting the most cost-effective therapy for treating male-factor infertility. It quantifies the number of germinal and white blood cells per mL of semen, which helps in determining the presence of disorders in spermatogenesis or genital tract infections. The test aids in the evaluation of sperm morphology abnormalities related to defects in sperm transport and fertilization processes, crucial steps for effective in vitro fertilization (IVF) treatment.
Special Instructions
Conventional semen analysis should be performed in conjunction with each strict criteria sperm morphology test. Specimen should be sent Monday through Friday, and the sperm count per mL is necessary for this test.
Limitations
Abnormalities in sperm morphology could result from improper specimen collection and handling, inappropriate test selection, or interfering substances. The presence of elevated germinal or white blood cells in semen may indicate underlying conditions whose clinical significance can vary.

Collection Instructions
If sperm concentration is <10 million/mL, centrifuge the specimen at 300 x G for 10 minutes before making slides. Label 2 frosted slides in pencil with the patient's first and last name and the date of specimen collection. Allow the semen to liquefy for 30 minutes. Place 10 mcL of liquefied semen on the label end of each slide, and evenly smear the specimen using a plain slide. Allow the smears to air dry for 15 minutes before placing both slides into 1 slide holder for shipment.
Patient Preparation
Patient should have 2 to 7 days of sexual abstinence at the time of semen collection for accurate results.
Causes for Rejection
All specimens will be evaluated at Mayo Clinic Laboratories for test suitability.
