Transcription Factor E3 (TFE3) Immunostain, Technical Component Only
Use
The TFE3 Immunostain test is particularly useful for assessing the expression of transcription factor E3, which is a member of the microphthalmia transcription factor (MiTF)/TFE family of helix-loop-helix transcription factors. Overexpression of TFE3 is observed in TFE translocation-associated renal cell carcinoma and alveolar soft part sarcoma. The immunostain test can help in confirming the diagnosis of these conditions by highlighting TFE3 expression in tissue samples.
Special Instructions
This test includes only the technical performance of the immunohistochemical (IHC) stain; no pathologist interpretation is provided. Technical component only stains should not be ordered with PATHC / Pathology Consultation, as they will be canceled if ordered together. For each additional IHC stain performed, an additional bill-only test ID will be reflected and charged. Clients wishing to access digital images must place the order for IHC stains electronically.
Limitations
This test does not include a pathologist's interpretation, which must be obtained separately. Age of a cut paraffin section can affect immunoreactivity, with stability thresholds varying widely among antigens. The charge of glass slides may be altered by environmental factors, impacting slide staining. Unsuitable glass slides can lead to inconsistent staining results due to poor slide surface chemistry. Best practices for positively charged slides include minimizing storage time post-unpackaging, limiting exposure to high humidity and heat, and minimizing exposure to plastics.

Collection Instructions
Formalin-fixed, paraffin-embedded tissue block or 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections should be 4 microns thick.
Storage Instructions
Attach the green 'Attention Pathology' address label (T498) and the pink Immunostain Technical Only label included in the kit to the outside of the transport container.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin fixed tissue, nonparaffin embedded tissue, noncharged slides, ProbeOn slides, Snowcoat slides.
