Tropomyosin Receptor Kinase (TRK) Immunostain, Technical Component Only
Use
This test is helpful in screening for neurotrophic tyrosine receptor kinase (NTRK) rearranged tumors. These are important to identify as patients with NTRK rearranged tumors have shown responses to TRK inhibitor therapy, making it significant in the management of driver-negative advanced malignancies such as secretory carcinoma, congenital infantile fibrosarcoma, and lipofibromatosis-like neural tumors.
Special Instructions
This test includes only the technical performance of the stain; no pathologist interpretation is provided. Do not order this test alongside PATHC / Pathology Consultation, as the technical component stains will be canceled in such cases. Any immunohistochemistry (IHC)/in situ hybridization (ISH) stain performed as a part of the PATHC will be carried out at the reviewing pathologist's discretion at an additional charge.
Limitations
The test does not include pathologist interpretation, only the technical performance of the stain. Stability of the antigen immunoreactivity may be affected by the age of a cut paraffin section, and there may be diagnostic confusion due to improper specimen collection and handling. Best practice dictates that paraffin sections be cut within 6 weeks, as stability thresholds can vary and are antigen dependent.

Collection Instructions
Preferred collection involves embedding tissue in formalin-fixed, paraffin-embedded tissue block or two unstained positively charged glass slides (25- x 75- x 1-mm) per test ordered with sections 4-microns thick.
