TTF41 (SPT24) + p40 Immunostain, Technical Component Only
Use
Thyroid transcription factor 1 (TTF1) aids in the classification of carcinomas of unknown origin, while p40 aids in the classification of carcinomas and lymphomas. TTF1 is often included in a panel to identify the primary site for carcinomas of unknown origin, due to its specificity for cells of thyroid or lung origin. The p40 antibody helps to distinguish squamous cell carcinomas from other non-small cell carcinomas, providing crucial diagnostic insights in complex cases involving undifferentiated tumor cells.
Special Instructions
This test includes only technical performance of the immunostains; no pathologist interpretation is provided. These technical component only stains should not be ordered with PATHC / Pathology Consultation, as the technical component stains will be canceled if ordered together. The ordering of PATHC allows for pathologists to determine the need for additional staining based on case specifics.
Limitations
The test involves immunohistochemistry on sections of paraffin-embedded tissue. Age of the paraffin section and environmental factors affecting glass slides may impact immunoreactivity. Immunoreactivity can decrease if sections have been cut more than six weeks prior to testing. Non-optimal conditions for glass slides can result in inconsistent staining.

Collection Instructions
Preferred specimens include formalin-fixed, paraffin-embedded tissue blocks, or 2 unstained, positively charged glass slides (25- x 75- x 1-mm) per test ordered; sections should be 4-microns thick.
Causes for Rejection
Wet/frozen tissue, cytology smears, nonformalin-fixed tissue, nonparaffin embedded tissue, noncharged slides, and ProbeOn/Snowcoat slides are causes for rejection.
