IGH::CCND1 (BCL1); t(11;14), paraffin
Use
FISH testing on paraffin-embedded cells for detection of IGH::CCND1 gene fusion. This fusion, associated with t(11;14), is observed in > 90% of mantle cell lymphomas and a subset of plasma cell neoplasms, follicular lymphoma, endometrial adenocarcinoma, and prolymphocytic leukemia.
Special Instructions
Tissues should be fixed in 10% neutral buffered formalin (NBF). Tissues preserved in B+ fixative or decalcified are usually not suitable for FISH. Assay requires 3 unstained slides minimum (two for each probe) and one H&E stained slide. If unstained slides are sent, cut FFPE tissue sections at 3-5 μm thickness, mounted on positively charged slides.
Limitations
Improper fixative, such as B+ fixative or decalcified tissues, may render the specimen unsuitable for FISH analysis. The test is limited by the quality and amount of tissue available. Specimens exposed to extreme temperatures or with insufficient tissue may be rejected.
