ROS1 Immunohistochemistry (IHC)
Use
ROS1 gene rearrangements occur in approximately 1%–2% of lung adenocarcinomas and are associated with response to the multi‑targeted tyrosine kinase inhibitor crizotinib. IHC for ROS1 protein expression is used as an alternate screening method for these rearrangements, with positive results recommended to be confirmed by ROS1 FISH studies.
Special Instructions
Any positive ROS1 IHC results must be confirmed by ROS1 FISH. Two levels of service are offered: Global and Tech‑Only (stain only).
Limitations
IHC provides screening with high sensitivity but relatively low specificity; false positives may occur and thus confirmatory testing (FISH) is recommended for any positive results.

Collection Instructions
Formalin‑fixed, paraffin‑embedded (FFPE) tissue block is preferred or one unbaked, unstained H&E slide (required) plus two to three positively charged unstained slides (cut at 4‑5 microns) per test/antibody.
Storage Instructions
Use cold pack for transport; cold pack must not contact specimen directly. NYS clients: provide date and time of collection.
