Chromosome Analysis, Products of Conception, with Reflex to Genomic Microarray

Use
This test is useful to ensure the highest chance of obtaining meaningful results from fetal specimens. It involves chromosome analysis of products of conception to detect chromosomal abnormalities that may be responsible for miscarriage or other adverse fetal outcomes. The test reflexes to genomic microarray analysis if the chromosome analysis is normal, improving detection of submicroscopic deletions or duplications that may not be visible through standard cytogenetic methods.
Special Instructions
Counseling and informed consent are recommended for genetic testing. New York State clients should note that testing is only approved for Chromosome Analysis, and reflex testing will be sent out. Informed consent is required with submission. A processing fee will be charged if the client cancels this procedure after the test has been set up or if the specimen integrity is inadequate to allow culture growth.
Limitations
The chromosome analysis involves culturing of living cells, which means that turnaround times are subject to variability. The standard cytogenetic methodology may not detect small rearrangements, low-level mosaicism, or submicroscopic deletions or duplications detectable by genomic microarray analysis. Additional cytogenetic testing by chromosome analysis or fluorescence in situ hybridization (FISH) may be needed for complete analysis of genomic imbalance.

Collection Instructions
Thaw media prior to tissue inoculation. Transport products of conception in a sterile, screw-top container filled with tissue transport medium. If specimen size is too large for a normal collection tube, a larger sterile container can be used.
Storage Instructions
Transport at room temperature if fresh; refrigerated also acceptable. Do not freeze or place in formalin.
Causes for Rejection
Frozen specimens. Intact fetus. Specimens in formalin. Specimens comprising maternal tissue (decidua) only. Autolyzed or contaminated specimens.
