Use
This test is used in the context of intrauterine fetal demise or stillbirth when further cytogenetic analysis is required. It's particularly relevant for pregnancy loss or termination in the presence of fetal anomalies, chromosomal abnormalities detected through conventional cytogenetic methods, multiple fetal losses of unknown etiology, or when products of conception specimens fail to grow in culture.
Special Instructions
Patients from New York will have their specimens sent to a New York state-approved laboratory. It's recommended that the test be ordered using the Cytogenetic test request form #43098. Informed consent is necessary for genetic testing, especially for NY patients.
Limitations
Certain genomic alterations may not be detected by this technology. These include CNVs below the resolution limit, sequence-level variants such as point mutations and indels, low-level mosaicism, balanced chromosomal rearrangements, and genomic imbalance in repetitive regions. Additional cytogenetic testing, such as chromosome analysis or FISH, is recommended for comprehensive evaluation. The test does not provide structural information of genomic imbalance and might miss some cases of tetraploidy.

Collection Instructions
Thaw media before tissue inoculation. Products of conception in a sterile container filled with culture transport medium. If no cytogenetics media is available, use plain RPMI, Hanks solution, sterile saline, or ringers. For autopsy, collect various tissue types such as facia lata, diaphragm, tendon, skin, etc.
Patient Preparation
Do not place the specimen in formalin. Skin or cord tissue should not be formalin-fixed.
Storage Instructions
Room temperature for fresh tissue or culture flask. Refrigerated or frozen is also acceptable.
Causes for Rejection
Intact fetus, specimens preserved in formalin, or contaminated specimens will be rejected.
