Use

Detect and quantitate HIV-1 in plasma with reflex to phenotype (at additional charge) if viral load is >1000 copies/mL.

Special Instructions

The test uses the Cobas® AmpliPrep/Cobas® TaqMan® HIV-1 Test, Version 2.0. A reflex test to Trofile® is indicated if the viral load exceeds 1000 copies/mL, incurring an additional charge. Ensure proper collection and processing of specimens to avoid delays. Separate frozen specimens are required for the Trofile® portion of the test. Stability varies depending on storage conditions; freezing is preferred for long-term storage.

Limitations

The assay range for the test is between 20 and 10,000,000 copies/mL, within which quantitation is considered reliable. Specimen integrity is critical, as hemolysis, use of improper tubes, or improper storage can lead to inaccurate results or rejection. The use of frozen PPT™ tubes can falsely elevate low viral loads, so plasma must be transferred to proper tubes prior to freezing.

Collection Instructions

Collect specimen in three lavender-top (EDTA) tubes or three PPT™ tubes. Centrifuge specimen within six hours of collection. Remove plasma, transfer to screw-cap polypropylene transport tubes, and freeze. Do not use green-top (heparin) tubes. Separate frozen samples are required for different tests.
