Use
This test is designed to detect elevated concentrations of plasminogen activator inhibitor 1 (PAI-1), which is thought to be a thrombotic risk factor. It helps in assessing the risk of thrombosis by measuring PAI-1 activity levels. However, the test is not a first-line diagnostic tool for inherited thrombotic or bleeding disorders. The assay assists in identifying the diurnal variation of PAI-1, with higher concentrations noted in the morning and decreased levels in the afternoon.
Special Instructions
Patient preparation involves specimen collection between 8 a.m. and 12 p.m. Centrifuge plasma and within 1 hour of draw, transfer 1.5 mL platelet-poor plasma to an ARUP Standard Transport Tube and freeze (Min: 1 mL). Adherence to specimen handling guidelines and storage instructions is critical for accurate results.
Limitations
Low concentrations of PAI-1 may not be accurately quantified by this assay, with concentrations below 2.0 IU/mL not being diagnostic of PAI-1 deficiency. The test result of less than 2.0 IU/mL may also be seen in normal individuals. Furthermore, the assay's accuracy is reliant on fasting samples and collection timing due to PAI-1's diurnal variation.
