Detection of the t(11;14)(q13;q32) translocation involving the IGH (immunoglobulin heavy chain) locus on chromosome 14 and CCND1 (cyclin D1, BCL1) on chromosome 11, characteristic of mantle cell lymphoma and found variably in other B‑cell neoplasms including B‑prolymphocytic leukemia, plasma cell leukemia, chronic lymphocytic leukemia, and multiple myeloma. This translocation results in cyclin D1 overexpression and is used to detect abnormal clones for diagnosis, prognosis, and response to therapy monitoring. Special Instructions
Specimen requirements include bone marrow aspirate (2 mL, sodium heparin green‑top) or peripheral blood (5 mL in sodium heparin). Minimum volumes are 1 mL bone marrow or 2 mL blood. Specimens must arrive at ambient temperature (20‑25 °C) within 24 hours; refrigerate if delayed. Frozen, clotted specimens, paraffin‑embedded tissue, or specimens in other anticoagulants are unacceptable. A cytogenetics/FISH requisition must accompany the specimen. Limitations
FISH analysis may be limited by low clone frequency or poor specimen quality. Detection of t(11;14) indicates presence of abnormal clone but a negative result does not exclude neoplastic disease.
