Use
Used to detect archived HIV‑1 drug resistance mutations in patients with virologic suppression or low‑level viremia (e.g., <1,000 copies/mL), when plasma RNA genotypic resistance testing may not be successful. The test captures historical resistance mutations in proviral DNA from host cells and may guide antiretroviral regimen modifications when plasma RNA is undetectable or insufficient.
Special Instructions
Collect 2 mL (minimum 0.6 mL) of whole blood in a lavender‑top EDTA tube. If ordering the combined resistance plus tropism panel (test code 94810), collect 3 mL (minimum 1.2 mL) whole blood. Proviral DNA testing is appropriate when routine RNA resistance testing fails due to low viral load.
Limitations
PCR primers amplify HIV‑1 subtype B and many major non‑B subtypes (A, C, BF, D, AE, AG, H), but uncommon non‑B subtypes or variants may not amplify. Clinical performance and utility are limited; some resistance mutations may be missed, while others may be archived and present without known clinical significance. Proviral DNA should not be considered a comprehensive archive; plasma RNA genotypes remain informative.
