Use
Chimerism analysis is essential for monitoring patients who have undergone allogeneic stem cell transplantation. This test is specifically used to assess the engraftment of donor cells in a bone marrow transplant recipient by analyzing sorted CD33+ cells. Accurate chimerism analysis is crucial for evaluating the success of the transplantation procedure, guiding further treatment decisions, and detecting potential complications such as graft rejection or graft-versus-host disease.
Special Instructions
Each posttransplant specimen and test ordered must have a unique ARUP accession number. Donor and recipient specimens must be obtained and genotyped before the transplant event occurs. Results such as recent white blood cell and differential counts should be included. For bone marrow specimens used in cell sorting, information on the cellularity of the patient's bone marrow is necessary.
Limitations
The test has a limit of detection of 2 percent of the minor cell population, meaning it may not detect low-level chimerism accurately. Sequence variations, although rare, can lead to diagnostic errors, potentially affecting the accuracy of chimerism analysis. Interpretative data must consider the clinical context, including the interval between transplantation and testing, which can impact result interpretation.
