Direct Coombs (Anti-Human Globulin)

Use
The Direct Coombs test is used to detect in vivo coating of red blood cells (RBCs) with immunoglobulin and complement degradation products on the patient's RBCs. This test is useful for antibody identification and evaluation of transfusion reactions, hemolytic disease of the fetus and newborn (HDFN), and autoimmune hemolytic anemia that may be associated with certain diseases or drugs.
Special Instructions
Do not freeze the sample. Transport 5 mL whole blood. Separator or gel tubes are unacceptable.
Limitations
The test may not precisely characterize the type of coating (e.g., IgG or C3d) if the reactivity is too weak. Interpretation requires consideration of clinical context.
