InSight: Prenatal Amnio Aneuploid (FISH) Testing for Chromosomes 13, 18, 21 and XY
Also known as: Direct Amnio FISH, Direct Prenatal FISH, InSight FISH, Prenatal Aneuploid, Prenatal FISH
Use
Fluorescence in situ hybridization (FISH), Prenatal aneuploid evaluation FISH targeting numerical changes in 13, 18, 21, X, Y. If specimen volume is too small, then direct FISH may not be performed and results may be obtained if cultured chromosome studies are ordered.
Special Instructions
InSight FISH testing must be performed concurrently with prenatal chromosome analysis or a microarray. Any pertinent medical findings should accompany the request for FISH. Abnormal results should have banded chromosome confirmation to ensure accuracy. It is critical to transfer the specimen to the cytogenetics laboratory immediately after collection and maintain it at room temperature. Do not freeze the sample, and refrigerate only if sterility is questioned or if the sample cannot be shipped within 24 hours.
Limitations
This FISH test does not detect molecular mutations below its resolution capacity and will not identify aneuploidies that are not specifically targeted by the FISH probes in this panel. Therefore, other chromosomal abnormalities or genetic mutations that do not affect chromosomes 13, 18, 21, or the sex chromosomes may not be detected with this test. Additionally, the test has not been cleared or approved by the Food and Drug Administration, which may impact its perceived reliability for certain clients.

Storage Instructions
Maintain specimen at room temperature and transport to the cytogenetics laboratory immediately. Do not freeze. Refrigerate if sterility is questioned or if sample cannot be shipped within 24 hours.
Causes for Rejection
Frozen amniotic fluid; wrong specimen type; low specimen volume; specimen in fixative; glass containers and/or rubber stoppered tubes (rubber is toxic to amniocytes); hypocellular specimen
