Limitations

The test may not detect variants located in regions outside the covered areas, such as noncoding or deeply intronic regions. Microsatellite and mosaics, along with genomic rearrangements like transposable element insertions, may not be reliably detected. Sequencing may be affected by allele drop-out or homopolymeric regions. Pseudogenes can interfere with detection in certain genes. Del/dup analysis of genes like PMS2 may be complicated by homologous pseudogenes like PMS2CL, which are not associated with Lynch syndrome. Translocations, certain rearrangements, and exact breakpoints of deletions or duplications may not be determined. Additionally, somatic variants are not targeted, and results can be affected by bone marrow transplantation.
