H3K27M by Immunohistochemistry
Also known as: H3K27M IHC
Use
This immunohistochemical stain detects the presence of the histone H3 K27M mutation in tissue specimens. The presence of the H3K27M mutation is diagnostic of diffuse midline glioma H3K27M-mutant, a clinically significant entity with prognostic implications, including poorer treatment response and adverse outcomes compared to wild-type tumors ([pubmed.ncbi.nlm.nih.gov](https://pubmed.ncbi.nlm.nih.gov/30647848/?utm_source=openai)).
Special Instructions
This test is performed as a stain-and-return technical service only. Ordering is recommended through the ARUP Immunohistochemistry Stain Offerings brochure. It is a Laboratory Developed Test (LDT) and is New York State Department of Health (DOH) approved ([ltd.aruplab.com](https://ltd.aruplab.com/index.php/Tests/Pub/3004370?utm_source=openai)).
Limitations
Specimen must be formalin-fixed, paraffin-embedded tissue or equivalent cell block with adequate representation; depleted specimens or non‑representative tissue types are unacceptable. Slides should be positively charged, 3–5 micron thick; if precut slides are sent, do not oven bake. Stain results are technical only; interpretation must be performed separately. Frozen specimens are unacceptable. Technical stain only — no pathologist interpretation is included ([ltd.aruplab.com](https://ltd.aruplab.com/index.php/Tests/Pub/3004370?utm_source=openai)).
