Use
Immunohistochemical staining for H3K27me3 (histone H3 lysine 27 trimethylation) is used as a technical stain-and-return service, typically to assist in diagnostic pathology. It is often helpful in evaluating tumor types such as malignant peripheral nerve sheath tumors or other neoplasms where loss or retention of H3K27me3 expression provides diagnostic insight. This test is performed laboratory‑developed test (LDT) and cannot be used for definitive mutation analysis, but serves as an adjunct immunohistochemical marker.
Special Instructions
This test is performed as a stain‑and‑return (technical) immunohistochemistry service only. Refer to the ARUP Immunohistochemistry Stain Offerings brochure for ordering recommendations. Components are provided for reference and cannot be ordered individually. For specimen transport, ARUP supply kit #47808 is highly recommended. If sending precut slides, do not oven bake.
Limitations
Only technical stain‑and‑return service is offered; interpretive evaluation must be done by the submitting facility. Frozen specimens are unacceptable. Slides or blocks must be represented; depleted or non‑representative tissue is unacceptable. Limited to immunohistochemical detection of H3K27me3 and does not assess other molecular alterations.

Collection Instructions
Formalin fix (10 percent neutral buffered formalin) and paraffin embed specimen (cells must be prepared into a cellblock). Transport tissue block or 5 unstained (3‑ to 5‑micron thick sections), positively charged slides in a tissue transport kit (ARUP supply #47808). If sending precut slides, do not oven bake.
Storage Instructions
Room temperature; Refrigerated acceptable. Ship in cooled container during summer months.
