MYCN (N-MYC) Gene Amplification by FISH

Use
This test is used for prognostic determination in individuals with neuroblastoma or medulloblastoma. It is diagnostic for the MYCN-amplified subgroup of spinal ependymoma. MYCN amplification, with a MYCN/CEP2 ratio of 2.0 or greater, is considered a feature of high-risk disease and an indicator of poor prognosis in neuroblastoma. Amplification is also seen in SHH-activated and group 4 medulloblastomas, with implications for prognosis and therapy response.
Special Instructions
Include surgical pathology report. If multiple specimens (blocks or slides) are sent to ARUP, they must be accompanied by a request for pathologist block/slide selection or individual orders, else they will be held until clarification is provided.
Limitations
This test was developed and its performance characteristics determined by ARUP Laboratories. It has not been cleared or approved by the US Food and Drug Administration. The test is performed in a CLIA certified laboratory and is intended for clinical purposes. Assay range and limit of detection were generated using normal and known positive cases respectively.

Collection Instructions
Formalin fix (10% neutral buffered formalin) and paraffin-embed tissue. Protect paraffin block from excessive heat.
Storage Instructions
Room temperature. Refrigeration is also acceptable. Ship in cooled container during summer months.
Causes for Rejection
Paraffin block with no tumor tissue remaining. Specimens fixed in any fixative other than 10% neutral buffered formalin. Decalcified specimens.
