BRAF by Immunohistochemistry

Use
This immunohistochemistry (IHC) stain is used to detect BRAF V600E protein expression in tissue or cell samples. As a Laboratory Developed Test (LDT) and stain-and-return service, it supports diagnostic evaluation in contexts such as oncology, allowing for indirect identification of the BRAF V600E variant via protein aberration. It is especially valuable as an initial screening tool to guide therapeutic decision-making where molecular mutation status is relevant.

Performed as a stain and return service only. Ordering recommendations point to the ARUP Immunohistochemistry Stain Offerings brochure. This test is New York State Department of Health approved. Compliance category indicates it is an LDT. The test is stain-and‐return only (slides returned to client).
Limitations
Requires formalin-fixed, paraffin-embedded tissue or cell block. Slides or blocks must be protected from excessive heat. If precut slides are sent, they should not be oven-baked. Nonrepresentative or depleted specimens are unacceptable. Frozen specimens are unacceptable. As an IHC stain, interpretation depends on tissue quality, and negative staining may not rule out non-V600E BRAF variants or low allele frequency mutations.

Collection Instructions
Collect tissue or cells; fix in 10% neutral buffered formalin and paraffin embed; protect paraffin block and/or slides from excessive heat; transport tissue block or slides in ARUP tissue transport kit (#47808); if sending precut slides, do not oven bake.
Storage Instructions
Room temperature or refrigerated; ship in cooled container during summer months.
Causes for Rejection
Nonrepresentative tissue type; depleted specimens.
