Special Stain, Periodic Acid‑Schiff (PAS), Fungus
Also known as: PASF SS
Use
This test is a histochemical special stain (Periodic Acid‑Schiff, PAS) used specifically for the detection of fungal elements in tissue specimens. As a stain-and‑return (technical) service, it allows pathologists to visualize polysaccharides, mucopolysaccharides, and fungal components which appear magenta after staining, aiding in diagnosis of fungal infections within tissues.
Special Instructions
Anatomic Pathology Test Request Form (#32960) is recommended for ordering. Specimens must be formalin‑fixed (10% neutral buffered formalin preferred) and paraffin‑embedded—or as a cell‑block. Transport tissue block or a minimum of one unstained, positively charged slide (3–5 micron sections) using a tissue transport kit (ARUP supply #47808). Submit electronic request or use form #32960. Protect specimens from excessive heat and ship in cooled container in summer.
Limitations
Cannot be used on frozen specimens; frozen is unacceptable. Unacceptable conditions include nonrepresentative tissue or depleted specimens. Specimens must be correctly fixed and embedded; otherwise staining quality may be compromised. As a technical stain service only, interpretive expertise is required by the submitting facility.
