Use
The Special Stain, Melanin Bleach test is utilized in histopathological evaluations to differentiate melanin pigmentation in tissue samples. This test is particularly significant in the assessment of pigmented lesions, as it aids in the distinction between melanin and other pigments or artifacts. Its application is crucial for accurate diagnosis and evaluation of melanoma and other pigmented skin conditions.
Special Instructions
An Anatomic Pathology Test Request Form (ARUP form #32960) is recommended to accompany the test order. Submit electronic request or use the ARUP Anatomic Pathology Form with an ARUP client number if electronic ordering is unavailable.
Limitations
The test has limitations in differentiating melanin from metal deposits and other non-melanin pigments that may not extract with the bleach. Specimens with nonrepresentative tissue types or those that are depleted may be unacceptable for this test. Moreover, frozen specimens are not stable, as they are considered unacceptable conditions for this methodology.

Collection Instructions
Formalin fix (10 percent neutral buffered formalin is preferred) and paraffin embed specimen. Cells must be prepared into a cellblock. Transport tissue block or 2 unstained (3- to 5-micron thick sections), positively charged slides in a tissue transport kit.
Storage Instructions
Room temperature or refrigerated. Ship in cooled container during summer months.
Causes for Rejection
Specimens submitted with nonrepresentative tissue type. Depleted specimens.
